Approved Research Project Scope

The approved project scope for this application is:
Given the complexity of genetic data, researchers often condense genetic predispositions for specific traits using polygenic scores (PGS). These scores leverage summary statistics obtained from genome-wide association studies (GWAS) to establish a weighted aggregate of all risky genetic variants that an individual carries. Despite the widespread utilization of PGS in empirical analyses, they do suffer from several limitations and deficiencies. First, we aim to show that the current methods of PGS construction lead to measurement error bias in the gene-environment studies. Second, in an effort to overcome these limitations, we aim to refine the current gene-enviornment models within a sample-splitting framework, and derive a new set of weights that can be used in polygenic scores and do not lead to measurement error bias in the gene-environment models. Finally, we plan to apply our new two-step approach and explore the intricate interplay between genes and the environment in shaping important health outcomes including heart attack, cancer, strokes, obesity, and blood pressure. Moreover, we aim to investigate the channels through which environment anf genes affect these outcomes. For that purpose we also plan to examine phenotypes concerning lifestyle such as accelerometer results and dietary habbits.
